Different proteins also have different tolerances to various proteases. Choosing a proper pH helps cells grow. It also stops protease activity. This prevents target protein degradation.<\/p>\n<p>In industrial production, pH is usually controlled at 4.5\u20135.5. Ammonia water is often added online during fermentation. It adjusts pH. A DO rebound means carbon sources are used up. Cells are hungry. You need to add carbon sources in time. If there is not enough oxygen, cell growth is limited. If DO is too high, toxic oxygen free radicals form in the broth. They kill yeast cells. DO is usually kept at 30% during fermentation. It is kept at 20% during the induction phase.<\/p>\n<h4>Bacterial Concentration<\/h4>\n<p>Studies look at fermentation performance with different initial yeast concentrations. They show initial concentration has a big impact on Pichia pastoris fermentation. Lower initial concentrations make cell growth faster. But they make protein expression less. Higher initial concentrations make substrate use slower. But they make protein concentration and yield higher. This is because yeast uses substrates for protein expression. It does not use them for cell growth.<\/p>\n<p>During the methanol induction phase, cells should reach a high concentration before induction. This helps them adapt to the methanol environment. It reduces methanol toxicity. Now, people stop feeding to start methanol induction.
